Tunable Multivalent Platform for Immune Recruitment to Lower Antigen Expressing Cancers

چکیده

Chemical immunotherapeutic strategies including Antibody Recruiting Molecules (ARMs - bivalent small molecules containing an antibody-binding domain (ABD) and a target-binding (TBD)) direct immune-mediated clearance of diseased cells. Anti-cancer ARM function relies on high tumor antigen valency, limiting against lower expressing tumors. To address this limitation, we report tunable multivalent immune recruitment (MIR) platform to amplify/stabilize antibody cells with valencies. An initial set polymeric ARMs (pARMs) were synthesized screened evaluate ABD/TBD copy number, ratio, steric occlusion specific induction. Most pARMs demonstrated simultaneous avidity binding anti-dinitrophenyl antibodies prostate-specific membrane antigens prostate cancer. Optimized mediated enhanced anti-cancer target compared analogous ARM.

T cells expressing chimeric antigen receptor promote immune tolerance.

Cellular therapies based on permanent genetic modification of conventional T cells have emerged as a promising strategy for cancer. However, it remains unknown if modification of T cell subsets, such as Tregs, could be useful in other settings, such as allograft transplantation. Here, we use a modular system based on a chimeric antigen receptor (CAR) that binds covalently modified mAbs to contr...

Diagnosis GLY120 Antigen for the Blood ‎and Breast Cancers Using Graphene ‎Nanosheet

The current study designed and simulated graphene nanosensors for detection of GLY120 tumor-associated carbohydrate antigens. Graphene is a two-dimensional nanosheet that offers a high surface-to-volume ratio and high mobility which increases its sensitivity as a graphene sensor over that of other nanoparticles. The current study simulated graphene sensors with and without GLY12...

A Viral Platform for Chemical Modification and Multivalent Display

The ability to chemically modify the surfaces of viruses and virus-like particles makes it possible to confer properties that make them potentially useful in biotechnology, nanotechnology and molecular electronics applications. RNA phages (e.g. MS2) have characteristics that make them suitable scaffolds to which a variety of substances could be chemically attached in definite geometric patterns...

Tunable Detectors for Artificial Immune Systems: From Model to Algorithm

Artificial immune systems (AIS) are often developed directly from textual descriptions of immune properties with little attempt to understand those properties free of an engineering domain or application. Here we present an example of how an AIS can be developed based on an in-depth investigation of an immune model. This model describes how the activation thresholds of immune cells can be tuned...
